The main topics of this course are:
- Membrane protein overexpression in host cells
- Membrane protein purification via IMAC (immobilzed metal ion affinity chromatography) and SEC (size exclusion chromatrography)
- Reconstitution of purified membrane proteins into lipid vesicles
- Functional analysis of reconstituted membrane proteins
Students will gain knowledge of all these methodologies and train their lab skills for working with E.coli or yeast cells in culture, protein chemistry using different chromatographic techniques, and suitable functional tests for their purified proteins. Quality control of the experiments will be determined using SDS-PAGE and Western Blots for proteins and spectrometry for cell growth and protein concentrations.
The main goal is to train the students' understanding of what, when, and why things are performed at certain points of an experiment and the interpretation of outcomes.
